If I'm building something that's supposed to be a "lightweight" track car, then ya, it really matters. 50-75 extra pounds hanging on the front of a car will make a noticeable difference in handling characteristics.

^ No way.

The bulk of the weight difference is in the trans. I would say between my DOHC ZC long block and lsv longblock there was a ~ 50 lb difference.

true if utilizing stock components for handling every pound makes a differince, if placing a substancially heavier motor in the car then what it was originally designed for is your intent then remember they are for the most part designed to be balanced. aftermarket sespension can help adjust the displacement but deppending on the over all goal of the vehical it may not matter. IE utilizing say a k20 in a 90 civic, all be it over kill but sick if you can keep it to the ground for track car. the substancial weight differince would make it hard as hell for a DD. So OP what would be your goal for the car. if its a dd, you should be fine B or D. its when you look at pure preformance applications that it comes down to 100lbs= aprox .01 of a second.
